产品信息

包含多种已获专利的质粒，此类质粒经适当的限制性内切酶消化完成后，产生的 12 条条带适合用作琼脂糖凝胶电泳分子量标准。消化后的 DNA 包含 100 bp 到 1,517 bp 的片段。500 和 1,000 bp 条带的强度更高，可用作参照带。提供每条 DNA 条带的近似质量（当上样量为 0.5 μg 时），可对分子量大小相近的样品进行粗略定量。

随附 1 管 6X 紫色凝胶上样染料，无 SDS。

NEB 还提供此 Ladder 的 Quick-Load 紫色染料版本。

• 注意事项

  1. 此 Ladder 不适用于对 DNA 进行精确定量，但是可对分子量大小相近的样品进行粗略定量。
  2. 建议使用上样缓冲液稀释并上样 0.5 μg 的 100 bp DNA Ladder。
  3. 所有片段均在 5´ 突出末端有 4 碱基，可使用 T4 多聚核苷酸激酶（T4 PNK，#M0201）对末端进行标记，或使用 DNA 聚合酶 I Klenow 片段（#M0210）补平（1）。使用 α-[³²P] dATP 或 α-[³²P] dTTP 进行补平反应。
  4. 100 bp DNA Ladder 可在 4℃ 环境中稳定贮存至少 3 个月。
  5. 长期贮存时，请贮存在 -20℃ 环境中。如需稀释样本，请使用 TE 或离子强度极低的其它缓冲液。如果使用 dH₂O 稀释，DNA 可能会变性。
  6. 由于技术的局限性，进行聚丙烯酰胺凝胶电泳时，可能会在 DNA Ladder 上看到一两条额外的条带。
