Monarch® RNA 纯化离心柱(500 µg) |

Monarch RNA 纯化离心柱(500 µg)是 Monarch RNA 纯化试剂盒(500 µg)(NEB #T2050)的组分,可用于从酶促反应,包括高效 RNA 合成反应中,纯化高达 500 µg 的 RNA。离心柱的设计没有使用固定环,确保不会出现缓冲液残留及交叉污染。RNA 可在低至 50 μl 的体积中洗脱,洗脱后可用于各种下游应用,包括 RT-PCR、NGS RNA 文库制备和转染。

Monarch RNA 纯化离心柱还提供 10 µg(NEB #T2037)和 50 µg(NEB #T2047)的结合能力。

Monarch® RNA 纯化离心柱(500 µg) |

1. 使用 HiScribe®; T7 快速高效 RNA 合成试剂盒(NEB #E2050)、1.5 – 1.8 µg DNA 模板,在 37℃ 下温育两小时,合成不同长度(0.6 – 8 kb)的 RNA 转录本。使用 Monarch RNA 纯化试剂盒(500 µg,T2050)对 40 µl 体外转录(IVT)产物进行纯化,并以 200 µl 的体积洗脱。使用 Nanodrop® 分光光度计测量 A260 计算出 RNA 产量,每个 IVT 反应的 RNA 产量范围为 268 – 425 µg。

2. 使用 SYBR Gold 染色的 1% 琼脂糖-TBE 凝胶,在 Millennium™ RNA Marker(Thermo Fisher Scientific)上评估 RNA 完整性(200 ng/泳道)。

Monarch® RNA 纯化离心柱(500 µg) |

使用 Monarch RNA 纯化试剂盒(500 µg,NEB #T2050)纯化 rRNA(来自 Sigma 公司 E. coli 16S 和 23S rRNA 标准品)。将 100 µl 无核酸酶水添加到离心柱,并在室温下温育 0、1、3 或 5 分钟,然后离心洗脱 RNA。使用 Trinean DropSense 16 测得的 A260 值计算出 RNA 回收率。温育五分钟所得产量最大。
