Next® RNA Mg++ 片段化模块 |

NEBNext® RNA Mg++ 片段化模块经过优化，在 94℃ 条件下与镁离子一起温育，可将 RNA 打断成小片段。片段大小由温育时间控制（图 1），并且通过添加 NEBNext 片段化终止液来结束反应。

Poly(A)+ mRNA（40 ng）纯化自通用人参考总 RNA（Stratagene），在 1X NEBNext RNA Mg++ 片段化反应缓冲液中，在 94℃ 条件下温育 1 – 5 分钟进行片段化。片段化反应在 1X NEBNext 片段化终止液中终止。样本用无核酶水中按 1:10 比例稀释，并通过 Bioanalyzer 2100 分析。
