NEBNext Ultra II DNA PCR-free 文库制备试剂盒包含不同起始量 DNA 文库制备所需的所有酶和缓冲液，以便在 Illumina 平台上进行二代测序，且不需要扩增步骤。快速、用户友好的流程仅需最少的手动操作时间，在无需 PCR 的情况下，就可获得优异的文库产量和 GC 覆盖度。

为方便起见，该试剂盒还提供 NEBNext 样品纯化磁珠（Beckman Coulter 的 SPRIselect 磁珠），可用于片段筛选和酶反应纯化步骤。

Next® Ultra™ II DNA PCR-free 文库制备试剂盒（含纯化磁珠） |

A：以 NA19240 基因组 DNA（Coriell Institute）为起始样本，使用 NEBNext Ultra II DNA PCR-free 文库制备试剂盒和 Illumina TruSeq® PCR-free 文库制备试剂盒分别制备 PCR-free 文库，350 bp 插入片段作为筛选标准。DNA 起始量为 1 µg。

B：分别使用 NEBNext Ultra II DNA PCR-free 文库制备试剂盒和 Roche Sequencing Kapa HyperPrep 文库制备试剂盒，结合 Covaris® 打断，制备 150-200 bp 插入片段的文库，不进行片段筛选。根据生产商推荐方法，NEBNext、Illumina 和 Kapa 试剂盒分别使用了 NEBNext 多样本接头引物试剂盒 1（Unique 双端 Index 引物，UMI 接头，适用于 DNA）、IDT for Illumina（TruSeq DNA UD Indexes）和 Kapa Dual-Indexed 接头。

Next® Ultra™ II DNA PCR-free 文库制备试剂盒（含纯化磁珠） |

相较于 Illumina DNA PCR-free 文库制备试剂盒，NEBNext Ultra II DNA PCR-free 文库制备试剂盒可实现更高的基因组覆盖度。根据生产商推荐方法，以 250 ng NA19240 基因组 DNA 为起始样本制备文库，使用 NovaSeq 6000 S4 v1.5 flow cell（PE 2×150 bp）测序。Read 去接头（fastp 0.20.0）后比对至端粒到端粒 CHM13 参考基因组（draft 1，bwa-mem 0.7.17），并标记重复（samblaster 0.1.24）。从每个 bam（sambamba view -s 0.6.8）中随机抽取 28 亿 Read。使用 mosdepth（v0.3.1，-F 772）评估过滤水平和初级覆盖度，并绘制常染色体或线粒体基因组图。图中标示出了预期覆盖度（num_reads*read length/genome size）。
