使用 NEBNext® ARTIC SARS-CoV-2 FS 文库制备试剂盒 – 含片段化酶(Illumina®)和 1.3 μl 的合并的扩增子的每个样品进行文库构建,使用 MiSeq® 仪器(2X75 bp)进行测序。为每个文库绘制每个扩增子的测序 reads。使用 seqtk(250,000 read pairs)向下抽样 reads 数,并使用 Bowtie 2 比对至 SARS-CoV-2 参考基因组(NCBI,NC_045512)。参考线表示 100 reads 的典型质量阈值。
