产品信息

BL21(DE3)源大肠杆菌(E. coli)化学感受态细胞 利用固相金属离子亲和层析(IMAC)分离含多聚组氨酸标签的重组蛋白时,目的蛋白经常被大量的大肠杆菌(E. coli)内源金属结合蛋白所污染。蛋白表达菌 NiCo21(DE3)经基因工程改造,使 IMAC 组分中的 E. coli 蛋白污染减少到最低:GlmS 在突变后消除了与 IMAC 树脂结合的能力,另外三种蛋白(SlyD、ArnA 和 Can)在加上重组标签后,能够通过几丁质亲和层析快速去除。

    A)在 NiCo21(DE3) E. coli 感受态细胞中表达谷氨酰 tRNA 合成酶(6-His),随后进行 Ni-NTA 纯化。将来自 Ni-NTA 柱的洗脱液(E)通过几丁质柱。目的蛋白在流动相(FT)中被洗脱,而 CBD 标签金属结合蛋白仍然保持与几丁质树脂(NEB #S6651)结合(B)。按照制造商推荐的条件进行纯化。B) 利用抗 CBD 抗体(NEB #E8034),通过 Western blot 检测污染物 ArnA、SlyD 和 Can。
