Next® Ultra™ DNA 文库制备试剂盒 |

用于 Illumina 测序平台的 NEBNext® Ultra™ DNA 文库制备试剂盒采用简化流程，包含用于二代测序文库制备的试剂，起始量为 5 ng – 1 µg。请注意，接头和引物不包含在试剂盒内，需要单独购买。

试剂盒内每个组分都通过严格的质量监控，每套试剂都经过文库制备，并在 Illumina 测序平台测序经过验证。

NEBNext Ultra™ DNA 文库制备试剂盒即便使用低起始量和复杂的样本，也可制备高产量文库Next® Ultra™ DNA 文库制备试剂盒 |
所有文库均以 5 ng 的 DNA 样本起始量进行制备，并在 Agilent Bioanalyzer® 上检测。
A：大肠杆菌基因组 DNA 文库。1：Ladder；2：使用 NEBNext DNA 文库制备预混液试剂盒（E6040）制备文库；3：使用 NEBNext Ultra DNA 文库制备试剂盒（E7370）制备文库。起始量为 5 ng，使用 NEBNext Ultra™ DNA 文库制备试剂盒制备的文库产量会显著提高。
B：FFPE 样本。基因组 DNA 提取自 66 岁男性的人类肺部肿瘤 10 年前的 FFPE 包埋块（Biochain, Inc.）。1：使用 NEBNext Ultra DNA 文库制备试剂盒（E7370）制备文库。2：起始 DNA（高度降解的 FFPE 基因组 DNA）。
Read 深度相关性显示 5 ng – 1 µg 的起始量具有一致的高覆盖度。
Next® Ultra™ DNA 文库制备试剂盒 |
使用不同起始量的人 IMR90 基因组 DNA（5、50 和 1000 ng）制备文库并进行 PCR 扩增。PCR-free 文库也使用 1000 ng 人 IMR90 基因组 DNA 进行制备。在 Galaxy 上分析人类基因组 10 kb 的序列覆盖度，使用 Microsoft® Excel® 计算 Pearson 相关系数（R2 值）。所有文库彼此高度相关，与 DNA 起始量和 PCR 扩增无关。所有 DNA 起始量都实现了高覆盖度。

起始 DNA：IMR90 基因组 DNA（人成纤维细胞）。

PF 错配率：所有与参考序列比对的碱基与参考序列的错配率。

重复百分比：标记为重复的匹配测序百分比。

嵌合体百分比：匹配到最大插入尺寸之外或两端匹配到不同染色体的 reads 的百分比。

这些数据说明即使低至 5 ng 的人类基因组 DNA 起始量，NEBNext Ultra DNA 文库制备试剂盒也能显示出优异性能。无论起始量多或少，错配率、重复百分比和嵌合体百分比都较低且相似。

批次质控

所提供的单独批次均通过额外功能验证。单个试剂通过标准酶活性和质控分析，并同时满足各单独组分页面列出的严格的附加质控标准
