NEBNext Ultra II FS DNA PCR-free 文库制备试剂盒包含的酶和缓冲液可将宽泛起始量范围的完整 DNA 样本转化为文库，以便在 Illumina 平台上进行二代测序，却无需扩增步骤。快速易用的流程支持可靠的酶促 DNA 片段化，可最大限度减少实际操作时间，无需 PCR 即可获得出色的文库产量和 GC 覆盖度。

Next® Ultra™ II FS DNA PCR-free 文库制备试剂盒 |

A：以人类细胞系 NA19240 基因组 DNA（Corielle 研究所）为起始样本，使用 NEBNext Ultra II FS DNA PCR-free 文库制备试剂盒和 Illumina PCR-free 文库制备试剂盒制备 PCR-free 文库，且以 350 bp 插入片段作为筛选标准。

B：使用酶片段化试剂盒 NEBNext Ultra II FS DNA PCR-free 文库制备试剂盒和 Roche Sequencing Kapa HyperPlus PCR-free 文库制备试剂盒制备 150 – 200 bp 插入片段的文库，不进行片段筛选。参照厂商推荐方案，将 NEBNext 多样本接头引物试剂盒 1 和 Kapa Dual-Indexed Adaptors 分别用于 NEBNext 试剂盒和 Kapa 试剂盒。

Next® Ultra™ II FS DNA PCR-free 文库制备试剂盒 |

相较于 Illumina DNA PCR-free 文库，NEBNext Ultra II FS DNA PCR-free 文库拥有更均匀的基因组覆盖度。两位实验人员参照厂商推荐的方案，以 250 ng NA19240 细胞系 DNA 为起始样本制备测序文库，并用 NovaSeq® 6000 S4 v1.5 flow cell（PE 2×150 bp）测序。Reads 去掉了接头（fastp 0.20.0），与端粒-到-端粒 CHM13 参考基因组（draft 1，bwa-mem 0.7.17）进行比对，并标记重复（samblaster 0.1.24）。从每个 bam（sambamba view -s 0.6.8）中随机取样 2.8B Reads。使用 mosdepth（v0.3.1，-F 772）评估初级覆盖度和过滤水平，并绘制常染色体或线粒体基因组图。预期覆盖度（num_reads*read length/genome size）如图所示。Illumina PCR-free 文库在不同实验人员之间存在明显差异；但 FS DNA PCR-free 文库在不同的实验人员之间高度一致。
