(粘性末端单位)
1 单位是指在 45℃ 条件下,当总反应体积为 50 µl 时,15 分钟能连接 50% 的 1 µg BstEII 酶切 λ DNA 的 12 碱基对粘性末端所需的酶量。反应条件

单位活性检测条件
1X Taq DNA 连接酶反应缓冲液和 DNA(20 µg/ml)。在 45℃ 下温育 15 分钟后,通过添加停止酶促反应的染料(50% 甘油,50 mM EDTA 和溴酚蓝)来终止反应,在 70℃ 下加热 10 分钟,然后上样到 0.7% 的琼脂糖凝胶上。由于存在连接酶,BstEII 酶切 λ DNA 的粘性末端将在 70℃ 热处理后保持在一起。

注意事项
- 反应条件:在 1X Taq DNA 连接酶缓冲液中以 45℃ 温育 DNA 和酶 15 分钟;或在热循环仪中温育 DNA 和酶,温育程序适用于 Barany 在《Genetic Disease Detection and DNA Amplification Using Cloned Thermostable Ligase》(利用克隆的热稳定连接酶进行遗传病检测和 DNA 扩增)(1991 年)中描述的反应。Proc. Natl. Acad. Sci. USA 88, 189-193. 用 50% 甘油、50 mM EDTA、溴酚蓝的混合物终止反应。
- 1X Taq DNA 连接酶反应缓冲液需要 NAD+ 作为辅因子。在 10X Taq DNA 连接酶反应缓冲液中已添加 NAD+;为延长 NAD+ 辅因子的半衰期,缓冲液应该 -80℃ 条件贮存。
- Taq DNA ligase will not ligate short 4-base overlaps (typical of restriction enzyme digests), while it efficiently ligates 12-base pair overhangs.
