NEBNext® Ultra™ RNA 非链特异性文库制备试剂盒包含用于制备 Illumina 测序的非链特异性 RNA 文库的试剂。请注意，接头、引物、rRNA 去除试剂和 poly(A) mRNA 分离试剂未随试剂盒提供，须单独购买。

试剂盒内每个组份都通过严格的质量监控，每套试剂都经过文库制备，并通过 Illumina 测序平台测序验证。

按照 NEBNext Ultra™ RNA 操作说明，使用 NEBNext Poly(A) mRNA 磁性分离模块（NEB #E7490），以 10 ng、100 ng 或 1 μg 总 RNA（通用人参考 RNA，Agilent #740000）作为文库起始量。文库在 Illumina 平台（GAIIx）上测序（单端 read）。对于每种起始量，在表达的转录本中都展现了良好的覆盖度。

    • 磁力架或磁力板（例如：NEBNext® 磁性分离架，NEB #S1515S；Alpaqua ® 96S 超磁性分离板，#A001322；或同等器材）
    • 80% 乙醇（新鲜配制）
    • Agencourt® AMPure® XP Beads（Beckman Coulter, Inc. #A63881）
    • 使用 NEBNext rRNA 去除试剂盒 – 人/小鼠/大鼠（NEB #E6310）时，需要 Agencourt RNAClean® XP（贝克曼库尔特公司，#A63987）
    • 0.1X TE，pH 8.0
